Title: Specificity and Energy Landscapes (SEL) guide the design of synthetic genome readers that govern cell fate choices

Abstract: We have developed high throughput methods that enable comprehensive exploration of the sequence specificities of DNA or RNA binding proteins and synthetic molecules. The specificity and binding energy landscapes (SELs) that emerge from such studies have yielded unanticipated insights into how proteins and small molecules search the genome for their cognate sites. These insights guided our efforts to engineer synthetic molecules that target genomic sites known to regulate genes that govern cell fate and cell state choices, including the onset of a range of neurodegenerative diseases and cancers.
